Immunohistochemistry (IHC) Tips
Deparaffinization and hydration:
For a sufficient reaction between antibody and antigen, deparaffinization steps should be:
- 1. Incubate at 60°C for 20 min;
- 2. Treat with xylene for 2 times, 10 min each;
- 3. For sections which are newly prepared, step 1 is better to be 60°C, 3-4 h.
In hydration steps there should be alcohol with gradiently decreased concentrations.
